Effector systems of cyclic AMP and progesterone have been shown to regulate granulosa cell proliferation in the preovulatory follicle. The purpose of the present study was to determine the potential role of both cyclic AMP and progesterone on morphological and functional interaction between granulosa cells and ovarian follicles. Granulosa cells obtained from small (< 2 mm) and large (2-4 mm) ovarian follicles of beef heifers were grown in primary culture for 48 hours. Granulosa cells obtained from medium-sized follicles expressed less of an inhibitory cell surface receptor for follicle-stimulating hormone than granulosa cells from small follicles. Cyclic AMP and progesterone also have a direct effect on granulosa cell proliferation, as evidenced by both light microscopy and tritiated thymidine incorporation. Small follicles and granulosa cells were incubated in the presence of progesterone, androstenedione, or 8-bromo cyclic AMP for 48 hours, with and without the cyclic AMP-dependent protein kinase inhibitor, KT-5720. Progesterone and androstenedione increased granulosa cell proliferation, and the increase was completely abolished by the addition of KT-5720. When small follicles were incubated with these hormones, in the absence or presence of KT-5720, progesterone and androstenedione decreased the inhibitory action of FSH. This is consistent with a synergistic interaction between cyclic AMP and progesterone on FSH-stimulated granulosa cell proliferation, since cyclic AMP activates FSH-stimulated protein kinase C.
